I live in jet ski heaven and your welcome to take a look.  Start on the Manatee river exit I-75 to Palmetto left (south) on 301/41 , Just before you cross the bridge into Bradeton cross over to the sand road to the left of the bridge. This is a great unofficial launch area.
Head out the river to your right.  The river will take you out and you will see Anna Maria Island straight ahead, Egmont Key a little to the North/East of Anna Maria and Fort Desoto to the North and East of Egmont.  
We usually head for Egmont key, circle the Island either direction.  We like to anchor near the light house and walk around a bit.  This is an uninhabited Island other than park rangers. Next you can zip over to Fort Desoto, You can follow a pretty straight path all the way through and pop out under the sky way. This route is well marked.  Finally back over to Anna Maria.  Look for the Piers on the North End.  Great meals can be had at both the Rod and Reel Pier and the Waterfront restaurant, (City Pier).  Drop a hook and watch your skis bob right in view.  A great way to wrap up the day is to then circle up to the north end of Anna Maria, cruise through the clothing optional sand bar and then stay really close to the tip of the Island to round it out into the Gulf of Mexico.  Follow the shore all the  way down until you re enter in Sarasota then back up the bay to the river.  
Now that is a perfect day.  
You could easily spend 3/4 days here because that is just one option.
